You need to re-install the MS 2005 C++ runtimes from this page: http://www.microsoft.com/en-us/download/details.aspx?id=26347

If you are using a 64-bit windows version you will still need to install the 32-Bit file "vcredist_x86.EXE" as Grasshopper is a 32-bit program.

It's possible that the installation of T-Splines might have changed the runtime files

Great, that was it!

...as Grasshopper is a 32-bit program.

Actually, to be pedantically accurate, Grasshopper is pure .NET code, and as such bit-depth agnostic. Which means that Grasshopper 'inherits' its bit-depth from whatever environment it is running in. Could be either Rhino4 (which is always 32-bit), Rhino5 32-bit or Rhino 64-bit. Only in the last case will Grasshopper act as a 64-bit program.

So why does Grasshopper need the 32-Bit version when running on a 64 bit machine running Rhino v5 64-Bit?

It shouldn't. The reason for the runtimes is because RhinoCommon requires them and Rhino5 (both 32 and 64) should install the required runtimes. The problem should only occur when running Grasshopper on Rhino4.
